Introduction
Over the years the hemocytometer has gone through a series of major developments creating a modern instrument that is more accurate and easier to use than its predecessors. Revvity’s disposable hemocytometer, known as CP2, is an easy-to-use enclosed chamber with counting grids for two samples. Because the CP2 slide is a single-use counting chamber, it reduces the potential issues with cross-contamination as well as improving efficiency and throughput.
How do I count cells with a manual Hemocytometer?
Manual cell counting with manual Hemocytometer
Step 1. Prepare the Hemocytometer
Clean hemocytometer and glass cover slip with 70% EtOH.
Step 2. Prepare Sample & Load Hemocytometer
Pipette 10 microliters of sample into the manual hemocytometer.
Step 3. Manually Count Cells in Sample
a. Place the hemocytometer under a microscope with a typical magnification of 100.
b. Focus both onto the grid pattern and the cell particles, and count the total number of cells found in 4 large corner squares
Step 4. Cell Calculations & Disposal of Hemocytometer
a. Multiply the dilution factor by the total number of cells, divide by the # of corner squares counted, and multiply by 10E4 to obtain cell concentration (cells/ml).
b. Clean hemocytometer and glass cover slip with 70% EtOH.

b. Dispose of the hemocytometer.
What Makes the Revvity Hemocytometer Different?
- Integrated design
- No cleaning or assembly of parts
- Two counting chambers per slide
- Disposable, so no washing biohazard
Details of the Revvity disposable Hemocytometer
Revvity’s disposable hemocytometer is an enclosed chamber with two ports for sample introduction. The chamber contains precisely spaced lines in a grid pattern. There are two counting chambers per slide for replicates.
The counting grid pattern is the Improved Neubauer, as in a common hemocytometer. The consistent pattern design allows the standard cell counting procedure to be followed.
