Simplifying chickpea seed sample homogenization and DNA extraction

Large seeds and nuts can be tough plant matrices resistant to consistent sample prep for downstream analysis. we describe a workflow useing the Omni Bead Ruptor™ 96, which allows for processing in both dry and buffered conditions, leading to high-integrity DNA at robust yields. Automation-enabled bead milling method allows matched throughput and compatibility with automated DNA extraction via the chemagic™ 360, offering a high-throughput solution that can significantly reduce labor-intensive, manual steps in the workflow. The approach utilizing sealed vessels rather than open tubes minimizes potential cross-contamination; hence, protocols described in this application note provides a consistent and efficient solution for downstream DNA-based applications, such as qPCR.
